Is DPPH Stable Free Radical a Good Scavenger for Oxygen Active Species?
نویسنده
چکیده
The reaction of 2,2-diphenyl-1-picrylhydrazyl free radical (DPPH) with active species of oxygen (hydrogen peroxide, tert-butyl peroxide, tert-butyl hydroperoxide, sodium peroxide, potassium superoxide, hydroxyl radical, tempo free radical, galvinoxyl free radical, Fremy’s salt, mchloroperbenzoic acid, potassium hydroxide, and sodium peroxynitrite) was reviewed and the new obtained results compared with previous literature data. It was shown that in all of these reactions the corresponding hydrazine DPPH-H (2,2-diphenyl-1-picrylhydrazine) is the major product, besides, in most of reactions, the nitro derivative of DPPH-H. The formation of the nitro derivative involves the previous formation of nitrogen dioxide, which can appear in the reaction mixture in two ways: via a radical attack on picryl moiety which releases the nitrogen dioxide or via a nucleophilic substitution of a nitro group, which releases the nitrite anion, which is subsequently oxidized to the nitrogen dioxide. Nitrogen dioxide reacts easily with DPPH to form the corresponding nitro derivative of DPPH-H. The scavenging activity of DPPH in terms of oxygen active species, which should yield oxy derivatives of DPPH, is very poor, only traces of these could be found.

The methanol extract of the dried ripe fruits of Alpinia rafflesiana was investigated for its DPPH free radical scavenger constituents. 2',3',4',6'-Tetrahydroxychalcone (7), which has never been isolated from natural sources was found to be most active as a DPPH free radical scavenger with the IC50 value of 55 microM. Other known compounds isolated from this species include 5,6-dehydrokawain (1...

tion, stable radicals have the advantage that their concentrations are readily and directly measurable. Among them a stable free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) was investigated as a reactive hydrogen acceptor and further found to be useful for the antioxidant determination. Since then DPPH has been mainly used to examine radical scavenging activity of antioxidative vitamins and po...

The phenolic compounds and the organic acids composition of the edible beefsteak fungus Fistulina hepatica was determined by HPLC/DAD and HPLC/UV, respectively. The results showed a profile composed by five phenolic compounds (caffeic, p-coumaric and ellagic acids, hyperoside and quercetin) and six organic acids (oxalic, aconitic, citric, malic, ascorbic and fumaric acids). The quantification o...
